Chronic Myeloid Leukemia (CML) cell models expressing drug-sensitive and drug-resistant forms of Bcr-Abl exposed to the action of tyrosine kinase inhibitors are analyzed by SR FTIR and Raman microspectroscopies to study the effects of drugs targeting abnormal Bcr-Abl associated tyrosine kinase activity characterizing those leukemic cells. The regions of phosphates (1290-900 cm-1) and amides (1700-1480 cm-1) ara analyzed and compared in the SR FTIR absorbance spectra of individual cells in order to identify IR changes related to the actions of drugs. The results of complementary investigations carried out in parallel samples will cross-validate the results of IR analysis. We expect to obtain cross-validated FTIR and Raman reference spectra of drug-sensitive and drug resistant CML cell models.
