The specific objectives of the research project are: i) the identification, as well as the phenotypic and functional characterization, of circulating myeloid cell subpopulations in tumor patients during treatment with immunomodulatory agents (specifically, anti-PD-1/anti-PD-L1inhibitors or Blinatumomab), to define the molecular and functional mechanisms at the basis of their pro-tumor action; ii) the development of pre-clinical animal models allowing the in vivo characterization and analysis of the interactions between tumor and myeloid cells, to validate experimentally the patient-derived observations and elucidate the sensitivity/resistance to the treatment with immunomodulatory agents.
Thanks to the ongoing collaboration among basic and clinical scientists and the innovative investigational approaches described in the present project, we will gain novel insights on cancer immune evasion mechanisms that restrain the efficacy of targeted immunotherapy, thus laying the ground for novel interventions to affect the natural history of many oncologic diseases and improve patients’ clinical outcome.
